Serum tau protein measurement

Blood samples were taken from the radial vein at 24 hours after birth (before EPO treatment) and 4, 8, and 12 days after EPO treatment. Blood samples were placed in a test tube (specifications: 5 mL, No A06277602; Hebi City Zhong Xing Medical Supplies, Hebi, China), maintained without stirring for 30 minutes at room temperature, and then centrifuged in a L-400 low-speed automatic balancing centrifuge (Hunan Xiangyi Laboratory Instrument Development, Changsha, Hunan Province, China) at 3,000 r/min for 15 minutes.Serum was removed into another test tube and placed in a freezer at ?70°C until use. To avoid any influence on the results, blood transfusions to the neonates were avoided before collecting blood samples. Serum tau protein was detected using an enzyme-linked immunosorbent assay. The kit was provided by the Shanghai HuDing Biological Science and Technology Co., Ltd.

PP2A and MPF enzymatic activity and VTG content

PP2A enzymatic activity was assayed according to the instruction of Serine/Threonine Phosphatase Assay System (Promega, V2460;Madison, Wisconsin, USA). Live oocytes were rinsed and homogenized

in ice-cold PSB to obtain homogenate. The homogenate was added into the provided spin columns with Sephadex beads to remove endogenous phosphates. The supernatant was taken for detecting total soluble protein concentration using a bicinchoninic acid assay (BCA kit;Beyotime, Shanghai,China). The reaction was initiated by adding 15 μL of reaction premixes containing 5 μL of 1 mM phosphopeptide and 10 μL of PPase-2A 5×reaction buffer. After reaction at 37 °C for 30 min,50 μL of stop solution was added to each well. The data was read at 630 nm. The activity levels of MPF and the contents of VTG were determined through the zebrafish MPF ELISA Kit (Mlbio, Shanghai, China) and the zebrafish VTG ELISA Kit (Huding, Shanghai, China), following the manufacturer’s protocol.

Serum biochemical, infammatory oxidative stress measurements 

Blood samples collected were used for serum biochemical examination, including fasting glucose, total cholesterol, triglyceride, low-density lipoprotein cholesterol (LDL-c), high density lipoprotein cholesterol (HDL-c) and creatinine level by full automatic biochemical analyzer. Fasting insulin and GLP-1 level were assessed using Rabbit Insulin ELISA Kit (Wuhan Huamei Biological Engineering Co., Ltd, Hubei Province, China) and Rabbit Glucagon Like Peptide-1 ELISA Kit (Shanghai Huding Biological Technology Co., Shanghai, China) according to the manufacturer’s instructions respectively. Serum oxidative stress related markers Lipid Peroxidation Malondialdehyde (MDA) and 8-hydroxy-2 deoxyguanosine (8-OHdG) were assessed by MDA Assay Kit (Nanjing Jianchen Bioengineering Institute, Jiangsu Province, China) and Rabbit 8-OHdG ELISA Kit (Shanghai Huding Biological Technology Co., Shanghai, China). Serum antioxidant enzyme superoxide Jianchen Bioengineering Institute, Jiangsu Province, China). And the infammation maker high-sensitivity C-reactive protein (hs-CRP) level was detected using Rabbit hs-CRP ELISA KIT (Wuhan Huamei Biological Engineering Co., Ltd, Hubei Province, China).

Materials and reagents

RAW264.7 cells were obtained from the cell bank of the Chinese Academy of Sciences(Shanghai,China). LPS from Escherichia coli O55:B5,3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT), fetal bovine serum, and hematoxylin and eosin (H&E) solutions were purchased from Sigma-Aldrich Trading Co., Ltd. (Shanghai,China). The SOD Assay Kit, Malondialdehyde (MDA) Assay Kit, Nitric Oxide (NO) Assay Kit and Nitric Oxide Synthase (NOS) Assay Kit were purchased from Jiancheng Bioengineering Institute (Nanjing, China).The Human Interleukin 6 (IL-6) ELISA Kit, Tumor Necrosis Factor-α(TNF-α) ELISA Kit and Inducible Nitric Oxide Synthase (iNOS) Assay Kit were purchased from Shanghai HuDing Biological Technology Co., Ltd.(Shanghai, China). Phosphate-buffered saline (PBS, pH 7.4) was purchased from Solarbio Science and Technology Co., Ltd. (Beijing, China).All other chemical reagents were of analytical grade. The ACE inhibitory peptides Leu-Tyr (LY), Arg-Ala-Leu-Pro (RALP) and Gly-His-Ser(GHS) were synthesized with purity over 95% by Synthgene Biological Technology Co., Ltd. (Shanghai, China).
